LY2928057 is a monoclonal antibody that targets ferritin. LY2928057 binds to ferritin and blocks its interaction with ferritin, leading to iron efflux, which results in elevated serum iron and transferrin saturation levels, as well as increased ferritin levels in monkeys and humans. [1]
Methods: Human pulmonary arterial smooth muscle cells (hPASMCs) were selected. Cells were pretreated with LY2928057 (1 μg/mL) for 1.5 hours, followed by stimulation with ferritin (1 μg/mL) for 2.5, 24, and 48 hours. RT-PCR, Western blot, ELISA, and immunofluorescence assays were used to detect the expression of ferroportin, ferritin, CD163, and IREB2.
Results: LY2928057 blocked the endocytic degradation of ferroportin caused by ferritin binding to it, thereby preventing cellular iron accumulation.[3]

The administration of LY2928057 (10-30 mg/kg) resulted in a dose-dependent increase in serum ferritin in cynomophores.[2]
