In vitro | Glutamate-5-kinase-IN-1 (compound 50) at a concentration of 40 μM, effectively inhibits G5K activity, achieving an IC50 value of 22.1 μM under conditions with 10 mM L-Glu and ATP. At concentrations of 5, 10, and 20 μM, this compound exhibits negligible cytotoxicity in HepG2 cells. Furthermore, at 120 μM, Glutamate-5-kinase-IN-1 significantly elevates the Km(app) for ATP, while the concentrations of L-Glu (150 mM, 300 mM, and 600 mM) and ATP (10 mM) remain constant. |
