Instructions
1. Solvent selection:
CY5 triethylamine salt is water-soluble and is usually dissolved in deionized water, distilled water or appropriate buffer solutions. The common concentration range is 1–10 µM, and the specific concentration can be adjusted according to experimental requirements.
2. Labeling reaction:
CY5 can be used to label antibodies, proteins, nucleic acids or other biomolecules. The common labeling method is to react with the target molecule through the active group. Mix the CY5 solution with the molecule to be labeled (such as an antibody or nucleic acid). The reaction conditions are generally carried out at room temperature and the reaction time is usually 1–2 hours.
3. Reaction termination and purification:
After the reaction, dialysis or gel filtration columns can be used to remove unbound dyes to ensure that the labeled molecules are pure and avoid background signals.
4. Detection and analysis:
Fluorescence microscopy: Use a fluorescence microscope to observe the labeled samples. The excitation wavelength of CY5 is 649 nm and the emission wavelength is 667 nm.
Flow cytometry: Fluorescence signals are analyzed by flow cytometry to detect the labeling of cells.
Fluorescence spectrophotometer: Detect the fluorescence intensity of the labeled sample, usually at 649 nm excitation and 667 nm emission.
Notes:
1) CY5 is light-sensitive and should be used away from strong light, especially during the labeling reaction.
2) When storing, it is recommended to keep CY5 in a dark place to prolong its fluorescence properties.
