| In vitro | CSF1R-IN-21 (compound 7e) at a concentration of 10 μM for 6 hours inhibits CSF-1R autophosphorylation and exhibits minimal cytotoxicity in the EOC 20 cell line according to cell viability assays [1]. At 50 μM for 4 hours, it shows good blood-brain barrier permeability in parallel artificial membrane permeability assays (PAMPA) [1]. The compound demonstrates good performance in plasma stability tests at 10 µM (0/30/120 min) [1]. In microsomal stability tests, CSF1R-IN-21 exhibits moderate stability at a concentration of 1 mM for 30 minutes [1]. In cell viability assays, the compound showed very low cytotoxic effects on the microglial cell line EOC 20 [1]. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
