| In vitro | CDK12-IN-9 (Compound 17) is a potent inhibitor of purified CDK12 kinase, with an IC₅₀ of 2.2 ± 0.74 nM. It also inhibits purified CDK13, CDK9, CDK7, and CDK2 kinases with IC₅₀ values of 13 nM, 466 nM, 1491 nM, and 551 nM, respectively. Furthermore, CDK12-IN-9 inhibits intracellular pSER2, a downstream marker of CDK12/13 activity, with an IC₅₀ of 17 ± 4.1 nM, and also inhibits pSER2 activity in whole blood, with an IC₅₀ of 209 ± 106 nM. |

For example, if the intended dosage is 10 mg/kg for animals weighing 20 g , with a dosing volume of 100 μL per animal, and a total of 10 animals are to be administered, using a formulation of

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
