| Kinase Assay | Boc-Val-Pro-Arg-MCA TFA participates in the activity assay of trypsin Operation steps 1. Add 100 μL of diluted Trypsin enzyme to a test tube containing 800 μL of 50 mM Tris-HCl buffer (pH 8.0). 2. Add 100 μL of 50 μM substrate Boc-Val-Pro-Arg-MCA TFA to start the reaction immediately. Then incubate at 55°C for 10 minutes. 3. After incubation, quickly add 1.5 mL of stop agent (methanol: n-butanol: distilled water = 35:30:35) to terminate the reaction. 4. Use a fluorescence spectrometer to measure the fluorescence intensity at an excitation wavelength of 380 nm and an emission wavelength of 450 nm. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
