Description
BChE-IN-4 is a highly potent inhibitor of the enzyme butyrylcholinesterase, capable of crossing the blood-brain barrier. It effectively reduces learning and memory impairments resulting from a deficiency in cholinergic activity in a mouse model. BChE-IN-4 holds promise for further investigation in the field of Alzheimer's disease research [1].
In vivo
BChE-IN-4 (compound 1) administered at doses of 10, 20, and 30 mg/kg effectively mitigates learning and memory impairments associated with cholinergic deficits in an Alzheimer's disease (AD) mouse model, without inducing adverse motor effects at 30 mg/kg. Studies on adult male Albino Swiss CD-1 mice with scopolamine-induced memory impairments revealed that BChE-IN-4 significantly enhances step-through latencies, indicating memory improvement. Additionally, in C57BL/6J mice (18-22 g), a 30 mg/kg dosage enhanced long-term memory retrieval by day 12 in the Barnes maze task, though it did not initially improve learning abilities or short-term memory retrieval by day 5. Further examination at a 30 mg/kg dosage, suspended in 1% Tween 80, confirmed that BChE-IN-4 does not affect locomotor activity nor cause motor deficits in rotarod tests, affirming its safety and efficacy in improving cognitive functions without impairing motor skills.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
