Description
ATR-IN-31 is a selective ATR kinase inhibitor with an IC50 of 7 nM. It does not significantly inhibit ATM kinase activity and can suppress ATR kinase activity at the functional level. ATR-IN-31 is capable of inhibiting the viability of prostate cancer cells and is applicable for research related to prostate cancer.

In vitro
ATR-IN-31 (compound M3) is a potent inhibitor of purified ATR kinase activity, with an IC50 of 7 nM, showing no significant inhibition on purified ATM kinase activity at IC50 >10000 nM. It effectively inhibits purified DNA-PK kinase activity with an IC50 of 179.6 nM. Additionally, ATR-IN-31 (compound M3) (0-50 μM; 120 h) significantly suppresses the viability of 22Rv1 human prostate cancer cells, achieving an IC50 of 0.21 μM after 120 hours of treatment. In human liver microsomes, ATR-IN-31 (compound M3) (2.0 nM) exhibits high stability with a half-life of 154.32 minutes, while in mouse liver microsomes, it demonstrates moderate stability with a half-life of 67.77 minutes.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
