Description
Antibacterialagent 332 is an inhibitor of Escherichia coli DsbA (EcDsbA). It reduces the swarming motility of E. coli without affecting bacterial growth and can be used in studies of bacterial infections (Escherichia coli).
In vitro
Antibacterial agent 332 (compound 17), in concentrations ranging from 0 to 100 mM, binds to Escherichia coli DsbA with a kinetic SPR K D of 2.2 μM and a steady-state SPR K D of 2.8 μM. When used at 50 μM with an injection interval of 180 seconds, its interaction with Escherichia coli DsbA yields an ITC K D value of 1.5 μM based on a single-site binding model. This compound inhibits the oxidation of native peptide substrates mediated by Escherichia coli DsbA in peptide oxidation assays with an IC 50 of 30 μM. Additionally, during ASMS screening, Antibacterial agent 332 demonstrated high enrichment rates, indicating the formation of stable complexes with Escherichia coli DsbA at room temperature over one hour.

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
