| Cell Research | 1. Solution preparation 1. Preparation of mother solution: Dissolve 6-ethyl-7-hydroxy-4-methylchromen-2-one in an appropriate solvent, such as PBS or DMSO, usually at a concentration of 1-10 mM. 2. Preparation of working solution: Dilute the mother solution with PBS/DMEM/H2O to a 1-10 μM working solution for later use. 2. Cell staining and imaging: 1. Add cell sample: Add the dissolved dye to the cell culture and incubate for a certain period of time (usually 30 minutes to 2 hours) so that the dye enters the cell and binds to the cell components. 2. Wash and observe under a microscope: After washing away the excess dye with PBS, observe the fluorescence signal of the cell under a fluorescence microscope. 3. Molecular probe experiment: 1. Labeling molecules or probes: React the dye with the target molecule (such as protein, DNA, etc.) for labeling. 2. Fluorescence detection: Detect the fluorescence signal of the target molecule through a fluorescence spectrometer or imaging system to analyze its properties or changes. IV. Drug screening experiment: 1. Treat cells with drugs: incubate the drugs with cell samples. 2. Add dye: after staining, observe the fluorescence changes of the drugs on cells. |
