| In vitro | Usage of Rhodamine 101 chloride [2]: (The following protocol is for reference only and should be appropriately adjusted according to your specific experiments.) 1. Preparation of Stock Solution Dissolve approximately 3-5 mg of Rhodamine 101 powder in 0.3 mL of DMSO, then add distilled water to achieve a final concentration of 1 mg/mL. Note: The stock solution can be stored stably for at least 3 weeks at 4°C in the dark. 2. Preparation of Working Solution (for combined staining with Mithramycin in ethanol-fixed cells) Dilute the stock solution with 0.15 M sodium chloride aqueous solution to achieve a Rhodamine 101 working solution concentration of 1.0 μg/mL. 3. Cell Staining 1) Preparation of ethanol-fixed cell samples: Fix the target cells (e.g., CHO cells) in 70% ethanol for at least 18 hours in advance, then centrifuge to remove the ethanol fixative. 2) Resuspend the centrifuged cells in an appropriate amount of the prepared Rhodamine 101-Mithramycin dual-dye working solution. 3) Incubate the cell-dye mixture at room temperature for at least 10 minutes to ensure thorough cell staining. 4. After incubation, analyze the stained cells using flow cytometry. The above information is based on published literature. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
