Description
PRRSV/CD163-IN-1 is an inhibitor targeting PRRSV and CD163 that blocks the interaction between PRRSV glycoproteins (GP2a, GP4) and the SRCR5 domain of CD163, suitable for research on porcine reproductive and respiratory syndrome (PRRS).
In vitro
Methods: In vitro protein-protein interaction assays were performed using 5 μM PRRSV/CD163-IN-1.
Results: It inhibited the protein-protein interaction (PPI) between PRRSV GP4 glycoprotein and the CD163-SRCR5 domain.
Methods: PAMs were treated with PRRSV/CD163-IN-1 for 24 h, and cytotoxicity was examined.
Results: Concentrations below 25 μM showed no obvious toxicity to PAMs, with an LC₅₀ of 81.7 μM [1].
Methods: PAMs were treated with 0, 5, 10, 15, 20 μM PRRSV/CD163-IN-1 for 4 h before PRRSV infection.
Results: It inhibited PRRSV infection of PAMs in a dose-dependent manner.
Methods: PAMs were treated with 15 μM PRRSV/CD163-IN-1 and infected with different genotypes of PRRSV.
Results: It significantly inhibited infection by both type Ⅰ and type Ⅱ PRRSV strains [1].
