Ponsegromab (PF 06946860) is a selective and potent humanized anti-GDF15 antibody inhibitor with IC₅₀ values of 0.123 nM, 0.053 nM, and 0.102 nM for human, rhesus macaque, and mouse targets, respectively. Ponsegromab exhibits anti-cachexia activity by binding to GDF15 to prevent its interaction with GFRAL, thereby disrupting GDF15/GFRAL-mediated signaling. Ponsegromab possesses potential anticancer activity and may be used to treat cancer patients with cancer anorexia-cachexia syndrome. Ponsegromab acts as a chemopreparative agent, increasing intracellular reactive oxygen species levels and reducing glutathione levels.

Targets & IC50
GDF15:0.1095 μg/mL (EC50), GDF15-GFRAL:0.5037 μg/mL
In vivo
Methods: Adult C57BL/6J mice were injected intraperitoneally with LPS (10 mg/kg) to induce sepsis. Prior to LPS injection, the anti-GDF15 neutralizing antibody Ponsegromab (30 μg per mouse) was administered via lateral ventricle injection.
Results: Ponsegromab significantly improved cognitive and memory function in septic mice (assessed via conditioned fear and novel object recognition tests), reduced hippocampal microglial activation and phagocytosis, and mitigated synaptic loss.[1]

Verified Activity
1. Immobilized Human GDF-15 Protein (His & HSA) can bind Ponsegromab. The EC50 is 3.834 ng/mL.
2. Immobilized Mouse GDF-15 Protein (His) can bind Ponsegromab. The EC50 is 5.7 ng/mL.
3. The binding of Human GDF-15 Protein (HSA & His) to the extracellular domain of biotinylated Human GFRAL Protein (Fc) can be blocked by Ponsegromab.
