Description
Myosin H Chain Fragment, Mouse is a peptide fragment derived from the α-myosin heavy chain that serves as a critical immunogenic determinant used to induce experimental autoimmune myocarditis (EAM) in murine models. Myosin H Chain Fragment, Mouse is applied in cardiovascular immunopathology research to elucidate the mechanisms underlying autoimmune-mediated cardiac inflammation and subsequent myocardial injury.
In vivo
Myosin H Chain Fragment, Mouse (150 μg) was emulsified with PBS/CFA (1 mg/mL, H37Ra) at a 1:1 ratio and then subcutaneously injected into mice, with two injections administered 7 days apart (on day 0 and day 7, respectively), which can induce experimental autoimmune myocarditis. [1]

  • Modeling Mechanism:

    Myosin H Chain Fragmen, as an autoantigen, contains pathogenic epitopes in its S2 subunit polymorphic region (amino acids 735-1032). After immunization, it activates autoreactive T cells and antibodies, leading to chronic inflammatory infiltration of the myocardium, myocardial cell necrosis, and ultimately myocarditis. The 838 and 955 amino acid polymorphisms in Myhc of BALB/c mice make them more susceptible to disease, while C57B/6 mice show significant resistance.

    Mice, BALB/c (susceptible), C57B/6 (resistant), Female, 4-6 weeks old

    Dosage and Administration Route:

    100 μg mouse cardiac myosin (BALB/c or C57B/6 origin) or rat α-myosin fragment (D0, D4 fragment, containing amino acids 735-1032), Emulsified in complete Freund's adjuvant (CFA), Subcutaneous injection

    Dosing Frequency and Duration Model:

    Immunised once on days 0 and 7

  • Validation:

    Histological markers: Chronic inflammatory cell infiltration (predominantly lymphocytes) and cardiomyocyte necrosis were observed in myocardial tissue; the incidence rate in BALB/c mice was significantly higher than in C57B/6 mice. Immunological markers: Anti-myocardial myosin-specific autoantibodies were detected in serum; autoreactive T cells targeting Myhc pathogenic epitopes could be isolated from the spleen.

*Precautions: All mice were sacrificed at predetermined time points after modeling, and their hearts, spleens, and serum were collected for testing.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
