Lipid TG4C, an ionizable cationic glycerolipid (pKa = 6.71), has been utilized for creating lipid nanoparticles (LNPs) designed to deliver mRNA in vivo. In mice, these LNPs containing lipid TG4C and mRNA encoding human erythropoietin (EPO) result in increased serum EPO levels. Additionally, inhalation of LNPs with lipid TG4C carrying mRNA for human hepatocyte growth factor (HGF) decreases levels of Il-1β, Il-6, and Tnf-α in bronchoalveolar lavage fluid (BALF) in a mouse model of pulmonary emphysema induced by porcine pancreatic elastase.
