| In vitro | LASSBio-2382 (Compound 17) effectively inhibits purified ROCK1 with an IC50 of 0.005 μM and ROCK2 with an IC50 of 0.003 μM in cell-free biochemical assays. The compound is insoluble in PAMPA-BBB experiments, preventing the determination of its blood-brain barrier permeability. At 30 μM for 24 hours, LASSBio-2382 reduces MDA-MB-231 cell viability by 28.8%. It inhibits MDA-MB-231 cell migration in a dose-dependent manner, lowering wound closure rates to less than 50% at 10 μM, under 25% at 30 μM, and showing precipitation at 100 μM. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
