In vitro | Enterocin is a very special structure with a unique and highly oxidized three-ring cage core skeleton with an α-pyrone unit and a benzyl group. The biosynthetic pathway of this compound begins with the assembly of the benzoic acid unit by a ligase, followed by seven iterative elongation steps mediated by type II polyketide synthases via a decarboxylative Claisen condensation with malonyl-CoA. Of particular interest is the oxidation of the C4 methylene group catalyzed by flavoproteins, which further triggers a Favorsky-like rearrangement and two intramolecular aldol condensations to build the three-ring cage skeleton. |
