Description
EG-011 is a first-in-class and potent activator of Wiskott-Aldrich syndrome protein (WASP) that relieves autoinhibition of WASP and robustly induces actin polymerization, EG-011 exhibits selective antitumor activity in lymphomas, supporting its use in cancer biology research to investigate cytoskeletal regulation, immune cell signaling, and WASP-driven tumor vulnerabilities.
In vitro
In lymphoma cell lines (OCI-LY-19, REC1), EG-011 induced dose-dependent cell death (20-55%) at concentrations of 500 nM to 2 μM over 72 hours. Treatment (500 nM-5 μM) triggered actin polymerization in sensitive cells within 4-24 hours, while showing no cytotoxicity towards normal PBMCs even at high concentrations (1-10 mM) [1].
In vivo
In female NOD-SCID mice bearing Mantle Cell Lymphoma (MCL REC-1) xenografts, intraperitoneal (i.p.) administration of EG-011 (200 mg/kg, 5 days per week) delayed tumor progression. At the conclusion of the study, tumor volume in the treated group was 2.2-fold smaller than that of the control group (P < 0.001), without observations of systemic toxicity [1].
