CN016

CN016 is a neuroprotective agent. It inhibits the elevation of pro-inflammatory cytokines G-CSF, GM-CSF, and IL-6 induced by Oxaliplatin. Additionally, CN016 suppresses the inflammatory response and the infiltration of immune cells into sensory neurons induced by Paclitaxel. Furthermore, CN016 protects neurons from neurotoxic damage induced by Paclitaxel and shields mice from Oxaliplatin-induced peripheral neuropathy.

CN016 (10-1000 nM) significantly protects primary DRG neurons in mice from neurotoxic damage induced by Paclitaxel.

CN016, administered via intraperitoneal injection at doses of 5-20 mg/kg one hour before each of the four alternate-day injections of Paclitaxel, significantly alleviates Paclitaxel-induced peripheral neuropathy in female C57BL/6J mice at doses of 10 and 20 mg/kg by restoring sensory function, protecting neural structure, and inhibiting neuroinflammation. Additionally, CN016 at 20 mg/kg (intraperitoneal injection) given one hour before each Oxaliplatin injection over a period of two weeks, effectively safeguards BDNF Val/Val mice from Oxaliplatin-induced peripheral neuropathy by modulating neuroinflammation pathways, maintaining sensory neuron integrity, and improving sensory function endpoints.
