| In vitro | Carbonic anhydrase-IN-2 (Compound 20) effectively inhibits the proliferation of various vancomycin-resistant enterococci (VRE) strains, including Enterococcus faecium (E. faecium) NR-31909 and Enterococcus faecalis (E. faecalis) NR-31971, with a MIC 50 of 184 nM and a MIC 90 of 3060 nM. It powerfully suppresses Enterococcus faecium α-carbonic anhydrase (Efα-CA) with a K i value of 31.6 nM and γ-carbonic anhydrase (Efγ-CA) with a K i value of 62.3 nM. Carbonic anhydrase-IN-2 demonstrates bacteriostatic activity against Enterococcus faecium NR-31909 over 24 hours. Against 12 intestinal symbiotic strains, including Lactobacillus, Bacteroides, and Bifidobacterium, it shows no inhibitory activity, with a MIC greater than 784 μM over 18-20 hours. Additionally, Carbonic anhydrase-IN-2 (0-128 μg/mL, 24 h) exhibits no significant cytotoxicity in African green monkey kidney cells, maintaining 95-100% cell viability. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
