Description
ALK-IN-24 is an orally active ALK inhibitor with an IC50 value of 1.7 nM. It also inhibits the insulin receptor kinase, with an IC50 value of 6 nM. ALK-IN-24 suppresses the proliferation of lung adenocarcinoma cells and hinders ALK-driven tumor growth in xenograft mouse models. It is applicable for research related to non-small cell lung cancer.
In vitro
ALK-IN-24 (Compound 29) effectively inhibits wild-type ALK and ALK L1196M mutant kinases in HTRF KinEASE-TK enzymatic assays, with IC50 values of 1.7 nM and 8.1 nM, respectively. Additionally, it inhibits insulin receptor kinase activity with an IC50 of 6 nM in the same assay. In cell-based proliferation assays, ALK-IN-24 suppresses the proliferation of H3122 cells with a CC50 of 36 nM and inhibits the growth of BaF3 cells expressing ALK L1196M with a CC50 of 120 nM.
In vivo
ALK-IN-24 (50 mg/kg; oral administration; once daily for 14 consecutive days) achieves an 86.3% tumor growth inhibition rate in SCID mice with H3122 xenografts.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
