AFM24 is a bispecific antibody with a TandAb structure expressed in CHO cells, simultaneously targeting the Epidermal Growth Factor Receptor (EGFR) and FcγRIIIA (CD16A). It consists of a huIgG1-scFv heavy chain and a λ light chain, with a predicted molecular weight of 196.34 kDa. As a novel innate immune cell engager, AFM24 binds with high affinity to CD16A on NK cells and macrophages, directing these effector cells to eliminate EGFR-positive tumor cells. Its mechanism primarily leverages enhanced antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP) to overcome limitations of conventional anti-EGFR therapies.
