AFM24 is a bispecific antibody with a TandAb structure expressed in CHO cells, simultaneously targeting the Epidermal Growth Factor Receptor (EGFR) and FcγRIIIA (CD16A). It consists of a huIgG1-scFv heavy chain and a λ light chain, with a predicted molecular weight of 196.34 kDa. As a novel innate immune cell engager, AFM24 binds with high affinity to CD16A on NK cells and macrophages, directing these effector cells to eliminate EGFR-positive tumor cells. Description

In vitro
AFM24 binds with high affinity to EGFR and CD16A, inducing potent NK cell-mediated cytotoxicity against EGFR-positive tumor cells. Its activity is independent of EGFR signaling, overcoming resistance driven by KRAS/BRAF mutations. It also promotes macrophage-mediated ADCP [1].
