| In vitro | sBADA TFA assay(for GAPDH quantification): Begin by washing S. venezuelae cells twice with pre-warmed fresh LB broth (30°C) and collect them using centrifugation at 7000 g for 2 minutes. Resuspend the cells in fresh LB broth (30°C) containing 1mM sBADA TFA and culture for 5 minutes. Treat the cells with 70% ice-cold ethanol and incubate them in an ice bath for 1 hour. Subsequently, wash the fixed cells twice with cold (4°C) 1x PBS by centrifugation, then resuspend them in 4°C 1x PBS prior to imaging. Guidelines (Following is our recommended protocol. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
