Description
sBADA TFA is an efficient green fluorescent dye, known as a sulfonated BODIPY-FL 3-amino-D-alanine. It is utilized for in situ labeling of peptidoglycan in bacterial cell walls. (λex=490 nm, λem=510 nm).
In vitro
sBADA TFA assay(for GAPDH quantification): Begin by washing S. venezuelae cells twice with pre-warmed fresh LB broth (30°C) and collect them using centrifugation at 7000 g for 2 minutes. Resuspend the cells in fresh LB broth (30°C) containing 1mM sBADA TFA and culture for 5 minutes. Treat the cells with 70% ice-cold ethanol and incubate them in an ice bath for 1 hour. Subsequently, wash the fixed cells twice with cold (4°C) 1x PBS by centrifugation, then resuspend them in 4°C 1x PBS prior to imaging. Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
