Pyroptosis, also known as cellular inflammatory necrosis, is a programmed necrosis; it is manifested by the continuous distention of cells until the cell membrane ruptures, leading to the release of cellular contents and thus activating a strong inflammatory response. According to the activation mechanism, it can be divided into caspase-1-dependent classical pathway and caspase-1-independent (caspase-4/5/11) non-classical pathway. caspase-1 and caspase-4/5/11 cleaves the Gasdermin-D (GSDMD) protein and release its N-terminal structural domain, which binds membrane lipids and perforates the cell membrane, leading to distension by changes in cellular osmotic pressure until the cell membrane ruptures. Both pathways simultaneously induce cleavage of IL-1β and IL-18 precursors to form mature IL-1β and IL-18, which are released into the extracellular space and cause an inflammatory response. Studying the mechanism of Pyroptosis helps researchers to understand its role and development in related diseases such as infectious diseases, neurological diseases, cardiovascular diseases, cancer and other diseases.
