| In vitro | Method: Splenocytes from DO11.10 and OT-II mice were stimulated with OVA(329-339) peptide (0.1-1 μM) under in vitro conditions to establish T cell lines. Using N-terminally and C-terminally truncated peptides as well as single amino acid substitution peptides (0.002-100 μM), T cell proliferation was measured by ³H-thymidine incorporation assay. Result: OVA(329-339) induced half-maximal proliferation in DO11.10 and OT-II cells at concentrations of 25 μM and 30 μM, respectively [1]. |
| In vivo | Method: BALB/c mice were subcutaneously immunized with OVA protein (10 μg, CFA adjuvant). Nine days later, T cells from draining lymph nodes were collected for clonal analysis. Result: Following in vivo immunization with 10 μg of OVA protein, OVA(329-337) was recognized as a T cell epitope, and OVA323-339 became a hotspot of the T cell response due to the presence of multiple overlapping epitopes [1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
