| In vitro | Multi-target kinase-IN-6 (Compound 12) at a concentration of 25 μM for 24 hours inhibits RyR2-mediated Ca 2+ leakage in HEK-293 RyR2 R-CEPIA1er cells. At 1.1 μM for 2 hours, it activates the sarcoplasmic reticulum Ca 2+ -ATPase (SERCA2a) in HL-1 cardiomyocytes. Additionally, at 2.3 nM for 10 minutes, it significantly enhances the ATPase activity of SERCA2a in sarcoplasmic reticulum microsomal vesicles isolated from adult mouse ventricles. The compound, at 10 μM for 30 minutes, inhibits NaV1.5-mediated Na + influx in CHO NaV1.5 Duo cells. Furthermore, at 10 μM for 24 hours, it does not exhibit significant cytotoxicity to HL-1 cardiomyocytes. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
