| Description | HLX316 (E-688) is an engineered human sialidase fusion protein that targets B7-H3 (CD276). HLX316 removes immunosuppressive sialoglycans from the tumor surface through enzymatic cleavage, thereby neutralizing Siglec-mediated immune evasion. HLX316 also specifically binds to B7-H3 on the surface of tumor cells, enabling precise tumor enrichment. HLX316 is suitable for use in studies of advanced solid tumors. |
| In vitro | The sialidase activity of HLX316 is more than 1,000 times higher than that of non-targeted sialidases, significantly enhancing ADCC and ADCP effects and strengthening antibody-mediated tumor cell killing.[1] |
| In vivo | In various mouse tumor models and the A375 humanized mouse model, HLX316 demonstrated more sustained desialylation activity in vivo. HLX316 demonstrated antitumor activity in multiple mouse models and showed superior efficacy compared to the control drug in the A375 humanized model. HLX316 was well tolerated in non-human primates with no significant toxicity, and the NOAEL was 150 mg/kg.[1] |
