Description
ELX-02 disulfate is a synthetic eukaryotic ribosome-specific glycoside (ERSG) that induces translation read-through of nonsense mutations, producing normally localized, full-length functional proteins. It can be used to study genetic diseases caused by nonsense mutations.
In vitro
ELX-02 disulfate (10/30 mg/kg; subcutaneous injection; twice weekly for 8 doses) accumulates in tissues in a dose-dependent manner [1].
ELX-02 disulfate administered at 10 mg/kg subcutaneously, both as a single dose and repeatedly (twice weekly for 7 doses), is rapidly absorbed into plasma. The plasma half-life of ELX-02 disulfate is 0.5 hours [1].
In CtnsY226X nonsense mutation mice, ELX-02 disulfate accumulates in renal tissue without apparent nephrotoxicity. Treatment with ELX-02 disulfate (10 mg/kg; twice weekly; for 3 weeks) reduced renal cystine accumulation in vivo [1].
In vivo
ELX-02 disulfate (100-400 μg/mL) can read through nonsense mutations in human cells [1].

Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSOTargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
