Cell Research | 1. Solution configuration 1. Mother liquor configuration: Dissolve Cy5-N3 in an appropriate solvent, usually DMSO, and formulate to the desired concentration (10 mM). 2. Working solution configuration: Dilute Cy5-N3 mother liquor with PBS/DMEM/H2O to the desired concentration, usually 1-10 µM. 2. Cell imaging Experimental steps: 1. Label cells: Add Cy5-N3 solution to the cell culture medium and incubate with the cells for a certain period of time, usually from 30 minutes to 1 hour (adjust according to the specific requirements of the experiment). 2. Rinse: After the incubation is complete, wash the cells through PBS buffer to remove excess dye. 3. Fluorescence microscopy imaging: Use a fluorescence microscope (excitation wavelength is 649 nm and emission wavelength is 670 nm) to observe the Cy5 marker in the cells and record the fluorescence image. III. Protein or nucleic acid labeling Experimental steps: 1. Reaction with protein or nucleic acid: Add Cy5-N3 to the protein or nucleic acid solution, the reaction time is usually 1 hour and the temperature is room temperature to ensure a labeling reaction with the target molecule. 2.
