| Description | CEMT is a carboxylesterase (CE) substrate and a ratiometric two-photon fluorescent reporter probe. Once hydrolyzed by CE, CEMT generates HMT, which serves as a sensor for mitochondrial pH. Upon activation by CE, CEMT exhibits ratiometric fluorescence changes in response to pH variations. It targets mitochondria and covalently binds to them, preventing leakage during acidification and enabling in situ imaging. |
| In vitro | CEMT (10 μM) in a cell-free 40 mM B-R buffer (pH 7.42, 37 °C) displays a linear ratiometric fluorescence response to carboxylesterase concentrations ranging from 0 to 15 μg/mL; its hydrolysis product, HMT, catalyzed by carboxylesterase (CE), shows a similar fluorescence response to pH values between 4.98 and 9.07. The intracellular hydrolysis product HMT of CEMT (10 μM; 20 min) exhibits strong mitochondrial colocalization in HepG2 cells, with minimal leakage even under acidic conditions (pH 4.98), confirming CEMT’s ability to anchor mitochondria for in situ imaging. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
