Description
CellTracker Orange CMRA Dye is a fluorescent dye used for cell imaging and tracking cell movement and location.
In vitro
CellTracker Orange CMRA Dye Cell Viability Assay: Begin by adding 5 μM dye to each sample, maintaining a temperature of 27-33℃ for 30 minutes to label the samples. Next, centrifuge the cells at 1000 g for 10 minutes, then resuspend the cell pellet in pre-warmed 10% iFCS RPMI at a concentration of 3*10^7 cells/mL. Incubate the cells again at 27-33℃ for another 30 minutes. Following this, centrifuge at 1000 g for 10 minutes and wash with pre-warmed PBS without Ca 2+ and Mg 2+. Centrifuge once more at 1000 g for 10 minutes and resuspend the cells in pre-warmed 10% iFCS RPMI. Finally, measure the fluorescence intensity with an excitation of 548 nm and emission of 576 nm. Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
