Description
Celltrack Deep Red is a fluorescent dye with a stable fluorescence signal that lasts for at least 72 hours. It is suitable for cell tracking and monitoring cell movement across multiple generations. Within a cell population, Celltrack Deep Red only transfers to daughter cells and does not transfer to adjacent cells (Ex/Em = 630/650 nm).
In vitro
Celltrack Deep Red is prepared initially as a stock solution dissolved in DMSO at a final concentration of 1 mM and stored in aliquots protected from light. For working solutions, it is diluted to a concentration of 250 nM-1 µM using a serum-free medium and pre-warmed to 37°C. For cell labeling, the protocol for suspension cells involves: 1) centrifuging to collect cells and discarding the supernatant, 2) gently resuspending cells in the pre-warmed working solution, 3) incubating under specific cell growth conditions for 15-45 minutes, 4) centrifuging and removing the dye, and 5) replacing with fresh medium, followed by fluorescence microscopy examination at Ex/Em = 630/650 nm. For adherent cells: 1) remove the medium and gently add the pre-warmed working solution, 2) incubate under specific conditions for 15-45 minutes, 3) remove the solution, 4) replace with fresh medium, and 5) examine with fluorescence microscopy at Ex/Em = 630/650 nm. Important notes include adjusting the concentration, incubation temperature, and time of the working solution based on specific requirements, minimizing dye concentration to maintain normal cell physiology and reduce artifacts, and avoiding buffers with amino or thiol groups.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
