1. Sample: RELA-Ko (Human) Cell Lysate at 30 μg Hela (Human) Cell Lysate at 30 μg
Primary: Anti-NFKB p65 (RELA) (TMAB-01228) at 1/1000 dilution
Secondary: IRDye800CW Goat Anti-Mouse IgG at 1/20000 dilution
Predicted band size: 65 kDa
Observed band size: 65 kDa
2. 25 μg total protein per Lane of various lysates probed with NFKB p65 monoclonal antibody, unconjugated (TMAB-01228) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at RT for 60 min.
3. 4% Paraformaldehyde-fixed Hela (H) cell; Triton X-100 at RT for 20 min; Antibody incubation with (NFKB p65) monoclonal Antibody, unconjugated (TMAB-01228) 1:50, 90 min at 37°C; followed by conjugated Goat Anti-Mouse IgG antibody (green) at 37°C for 90 min, DAPI (blue) was used to stain the cell nucleus. PBS instead of the primary antibody was used as the blank control.

Paraformaldehyde-fixed, paraffin embedded Mouse Spleen; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (TMAB-01228) at 1:200 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
5. Paraformaldehyde-fixed, paraffin embedded Rat Spleen; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (TMAB-01228) at 1:200 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
6. Paraformaldehyde-fixed, paraffin embedded Human Spleen;Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (ascites of TMAB-01228) 1:1500 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
7. Paraformaldehyde-fixed, paraffin embedded Human Tonsil;Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (ascites of TMAB-01228) 1:1500 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.

Paraformaldehyde-fixed, paraffin embedded Human Liver Cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (TMAB-01228) at 1:200 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.

Paraformaldehyde-fixed, paraffin embedded Human Colon Cancer; Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Antibody incubation with NFKB p65 Monoclonal Antibody, Unconjugated (TMAB-01228) at 1:200 overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
10. Research BackgroundNF-kappa-B is a ubiquitous transcription factor involved in several biological processes. It is held in the cytoplasm in an inactive state by specific inhibitors. Upon degradation of the inhibitor, NF-kappa-B moves to the nucleus and activates transcription of specific genes. NF-kappa-B is composed of NFKB1 or NFKB2 bound to either REL, RELA, or RELB. The most abundant form of NF-kappa-B is NFKB1 complexed with the product of this gene, RELA. Four transcript variants encoding different isoforms have been found for this gene.
