Anti-DLL3 Antibody (anti-DLL3 arm derived from AMG-757) is a humanized antibody arm derived from the half-life extended bispecific T-cell engager (BiTE) AMG-757. This product specifically targets Delta-like ligand 3 (DLL3), a Notch pathway ligand highly expressed in small-cell lung cancer (SCLC) and other neuroendocrine tumors but minimally expressed in normal tissues. It binds to the extracellular domain of DLL3 with picomolar (pM) affinity. In a bispecific format, it facilitates the redirection of T cells to DLL3-positive tumor cells, inducing T-cell activation and the release of cytotoxic granules to exert potent anti-tumor effects.

Description

In vitro
The anti-DLL3 arm binds human DLL3 with high affinity (KD ≈ 0.64 nM). In T-cell and SCLC co-culture assays, the antibody redirects T cells, leading to robust activation (CD25/CD69 upregulation) and secretion of IFN-γ and TNF-α, which results in potent, dose-dependent lysis of DLL3-expressing tumor cells [1].
