| Cell Research | Instructions 1. Prepare a culture medium containing Ac4ManNDAz (usually add a specific concentration of Ac4ManNDAz, such as 20–100 µM, adjusted according to experimental requirements). 2. Expose cells to the culture medium containing Ac4ManNDAz and incubate for a certain period of time (usually 24–48 hours) to allow the probe to be incorporated into the glycoprotein synthesis pathway. 3. UV cross-linking: Collect cells treated with Ac4ManNDAz and wash with PBS or appropriate buffer; expose cells to UV light of a specific wavelength (usually 365 nm) for an irradiation time optimized according to the experiment (e.g. 5–15 minutes); after cross-linking is completed, stop UV irradiation immediately to avoid nonspecific damage. 4. Subsequent analysis: a. Lyse cells: Extract cell lysate with lysis buffer (e.g. RIPA buffer). b. Detect glycoprotein interactions: Isolate and identify glycoproteins and their ligands incorporated with Ac4ManNDAz by co-immunoprecipitation, mass spectrometry or other methods. c. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
