| In vitro | This protocol outlines our suggested procedure and should be customized to suit your specific requirements. Enzyme activity (such as with rabbit osteoclasts): 1. Incubate purified, differentiated cells in medium for 48 hours (adjust time based on the cell type). 2. Transfer treated cells to 4.5 mL of lysis buffer. 3. Divide each lysate into three parts and incubate with 40 μM Ac-VEID-AMC at 37°C for 1 hour. 4. Measure the cleavage of fluorescent substrates, with excitation at 340-360 nm and emission at 440-460 nm. |
