| In vitro | SHP2-IN-46 (Compound 27) effectively inhibits the proliferation of cancer cells A549, MIA PaCa-2, and HepG2, with IC 50 values of 0.24 μM, 0.16 μM, and 0.09 μM, respectively, while showing low cytotoxicity in non-cancerous Vero cells, with an IC 50 value of 42.79 μM. At a concentration of 0.5 μM for 24 hours, SHP2-IN-46 induces S phase cell cycle arrest in HepG2 cells, thereby inhibiting cell proliferation. The compound demonstrates good metabolic stability in human liver microsomes, with a half-life (T 1/2) of 40.63 minutes and intrinsic clearance (CL int) of 34.11 μL/min/mg protein. SHP2-IN-46 also inhibits SHP2 enzyme activity, with an IC 50 value of 11.76 μM in a concentration range of 0.001-100 μM over 90 minutes. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
