eGFP-mRNA is an mRNA molecule capped at the 5' end, polyadenylated at the 3' end, and modified with N1-Me-Pseudo UTP, mimicking the processed mature mRNA found in eukaryotes. The mRNA is capped co-transcriptionally using 3'-O-Me-GAG, forming a Cap1 structure which enhances mRNA stability and translation efficiency. The incorporation of the modified nucleotide N1-Me-Pseudo UTP reduces innate immune stimulation and increases mRNA stability. The addition of the poly(A) tail further stabilizes the mRNA and enhances its translation initiation efficiency.

In vitro | eGFP mRNA-LNP (1 μg/mL, 24 hours) can be efficiently translated into EGFP protein in HeLa cells, but exhibits very low translation efficiency in macrophages[1]. eGFP mRNA-LNP (0.25–1 μg/mL, 6–48 hours) showed dose- and time-dependent transfection efficiency in human primary airway epithelial cells and lung fibroblasts. At low doses (0.25 μg/mL), transfection peaked at 24 hours. At high doses (1 μg/mL), transfection efficiency was nearly the same between 24 and 48 hours[2]. |
In vivo | In vivo imaging of C57Bl/6J mice 16 hours after a single intravenous injection of eGFP mRNA-LNP (2 mg/kg) revealed that eGFP mRNA was only translated into protein in hepatocytes, with no significant translation signals observed in Kupffer cells (KC). |
