Description
CVN14 is an efficient and selective CD38 inhibitor that can penetrate the blood-brain barrier, with an IC50 of 19 nM in humans and 2.4 nM in mice. It binds non-competitively with CD38 and ADPR to form a complex, thereby inhibiting CD38 enzyme activity. CVN14 is applicable for research into neurodegenerative diseases.
In vitro
CVN14 is a potent inhibitor of human CD38, with an IC50 of 19 nM, and it also inhibits mouse CD38 with an IC50 of 2.4 nM. In CHO-hCD38 cells, CVN14 effectively inhibits human CD38 activity, exhibiting an IC50 of 19 nM. The compound binds tightly to human CD38, with a dissociation constant (Kd) of 3.6 nM and a dissociation half-life of 143 minutes. Additionally, CVN14 demonstrates good in vitro permeability across MDR1-MDCK cell monolayers, with an efflux ratio of 0.90.
In vivo
The compound CVN14 (10 mg/kg; oral; single dose) demonstrates significant brain distribution, with a Kp uu,brain value exceeding 0.3.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
