| Verified Activity | 1. Anti-PPIL1 mouse monoclonal antibody at 1:500 dilution. -Lane A: U87MG Whole Cell Lysate. -Lane B: K562 Whole Cell Lysate. -Lane C: Hela Whole Cell lysate. -Lysates/proteins at 30 μg per lane. -Secondary -Goat Anti-Mouse IgG H&L (Dylight800) at 1/15000 dilution. -Developed using the Odyssey technique. -Performed under reducing conditions. -Predicted band size:18 kDa. -Observed band size:19 kDa(We are unsure as to the identity of these extra bands.) 2. Flow cytometric analysis of Human PPIL1 expression on MCF-7 cells. The cells were treated according to manufacturer’s manual (BD Pharmingen™ Cat. No. 554714), stained with purified anti-Human PPIL1, then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells. |

| Research Background | PPIL1 is a member of the cyclophilin family. Cyclophilins are well conserved and ubiquitous. Members of cyclophilin family take a significant part in protein folding, immunosuppression by cyclosporin A, and infection of HIV-1 virions. PPIL1 is a peptidylprolyl isomerase(PPIase). It increases the folding of proteins and catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. PPIL1 is involved in proliferation of cancer cells through modulation of phosphorylation of stathmin. It is a novel molecular target for colon-cancer therapy. |
