1. Paraformaldehyde-fixed, paraffin embedded (rat kidney); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15 min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30 min; Antibody incubation with (MSS1) Polyclonal Antibody, Unconjugated (TMAB-06852) at 1: 200 overnight at 4°C, followed by operating according to SP Kit (Rabbit) instructions and DAB staining.

The GTPBP3 is the mitochondrial GTPase evolutionarily conserved from bacteria to mammals. GTPBP3 is ubiquitously expressed in various tissues. GTPBP3 localizes in mitochondrion. The human GTPBP3 is a deafness associated homolog of yeast MSS1. The allelic variants in GTPBP3 could modulate the phenotypic manifestation of human mitochondrial A1555G mutation. GTPBP3 may be involved in the 5 carboxymethylaminomethyl modification (mnm(5)s(2)U34) of the wobble uridine base in mitochondrial tRNAs.
