Verified Activity | 1. Western Blot -Positive WB detected in: Hela whole cell lysate, Jurkat whole cell lysate, HepG2 whole cell lysate, HL60 whole cell lysate, U87 whole cell lysate -All lanes: FAS antibody at 1:2000 -Secondary: Goat polyclonal to rabbit IgG at 1/50000 dilution -Predicted band size: 38, 12, 10, 17, 15, 36, 25 kDa -Observed band size: 45 kDa 2. IHC image of TMAH-00419 diluted at 1:100 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB. 3. IHC image of TMAH-00419 diluted at 1:100 and staining in paraffin-embedded human colon cancer performed on a Leica BondTM system. Research Background | Receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. The secreted isoforms 2 to 6 block apoptosis (in vitro). |
