Hoechst 33258 is a blue fluorescent dye that can penetrate the cell membrane and specifically embed into the AT enriched region of DNA double stranded DNA. It has low toxicity to cells and is commonly used for nuclear staining. When Hoechst 33258 is not bound to DNA, the fluorescence intensity emitted by the free state Hoechst 33258 is extremely low, almost no fluorescence (maximum excitation wavelength is 346 nm, maximum emission wavelength is 460 nm); When Hoechst 33258 is embedded into double stranded DNA, its fluorescence characteristics change, and it can emit bright blue fluorescence upon excitation (maximum excitation wavelength of 352 nm, maximum emission wavelength of 461 nm), and the fluorescence intensity increases with the increase of DNA content, making it easier for quantitative analysis. Hoechst 33258 is commonly used for detecting cell apoptosis, and after staining, the staining results can be observed under a fluorescence microscope or detected by flow cytometry.
