Betaine reacts with Reinecke’s salt under strongly acidic conditions to form a precipitate. The precipitate is dissolved in acetone to produce a red solution, which shows a characteristic absorption peak at 525 nm. By measuring the absorbance at 525 nm, the betaine content in the sample can be calculated.

I. Preparation of Lab Instruments
Analytical balance, centrifuge, visible spectrophotometer, 1 mL glass cuvette, methanol, ether, hydrochloric acid, and distilled water.
II. Pre-preparation
CB0034S-ES (provided by the user)
Add 40 mL methanol and 10 mL distilled water, mix well.
CB0034S-Standard
Add 1 mL distilled water immediately before use and dissolve thoroughly.
CB0034S-A
Prepare fresh according to the required amount before use.
Add 4 mL distilled water to each vial to dissolve, then add 78 μL concentrated hydrochloric acid to adjust the pH to 1.
CB0034S-B (provided by the user)
Mix 14.85 mL anhydrous ether with 0.15 mL distilled water.
CB0034S-C (provided by the user)
Highly volatile; prepare as needed according to usage.
Acetone : distilled water = 7 : 3.
III. Extraction of Betaine
Weigh approximately 0.2 g of the dried sample passed through a 40-mesh sieve, add 1 mL of extraction solution, and extract at 60 °C for 30 min with continuous shaking. Centrifuge at 10,000 g and 25 °C for 15 min, then collect the supernatant.
IV. Assay Procedure

| Mix thoroughly, react at 4 °C for 2 h, centrifuge at 8000 g, 25 °C for 15 min, and discard the supernatant. | ||

| Centrifuge at 8000 rpm, 25 °C for 10 min, then place in a fume hood to allow CB0034S-B to naturally evaporate completely. | ||

| Shake to fully dissolve the precipitate. Using a 1 mL glass cuvette, zero with CB0034S-C, and record the absorbance at 525 nm (A525) for both the standard and test samples. | ||

V. Calculation
Betaine content (mg/g) = A525 sample tube ÷ A525 standard tube × Cstd × V1 ÷ (W × V2 ÷ V3) = A525 of sample tube ÷ A525 of standard tube ÷ W
Note:
Cstd:concentration of the standard tube, 1mg/mL;
V1: Volume of the standard, 0.25 mL;
V2: Volume of the sample in the reaction, 0.25 mL;
V3: Volume of extraction solvent added, 1 mL;
W: Sample weight, g.
1.When preparing CB0034S-A, the pH must be strictly controlled at 1; otherwise, the reaction will be incomplete. After preparation, it is stable at 4°C for only a few hours and should be used as soon as possible.
2.CB0034S-B and CB0034S-C can cause some respiratory irritation. Please take appropriate protective measures.
