BNC-101 is a humanized monoclonal antibody targeting Leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5/GPR49). LGR5 is a key component of the Wnt signaling pathway and a critical marker of cancer stem cells (CSCs) in various malignancies, particularly colorectal and pancreatic cancers. BNC-101 binds specifically to the extracellular domain of LGR5 with high affinity, inhibiting the abnormal activation of the Wnt/beta-catenin signaling pathway by blocking the interaction between LGR5 and its ligands, R-spondins. This antibody is designed to reduce tumor tumorigenicity, recurrence, and metastatic potential by targeting and eliminating LGR5-positive cancer stem cells.
