Verified Activity
1. Immunofluorescence staining of rat TNFα in rat spleen cells. Cells were fixed with 4% PFA,blocked with 10% serum, and incubated with rabbit anti-rat TNFα monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Cell membrane.
2. Anti-TNFalpha rabbit monoclonal antibody at 1:500 dilution.
-Lane A: Hela Whole Cell Lysate.
-Lane B: Rat spleen tissue Lysate.
-Lane C: Rat brain tissue lysate.
-Lysates/proteins at 30 μg per lane.
-Secondary
-Goat Anti-Rabbit IgG (H+L)/HRP at 1/10000 dilution.
-Developed using the ECL technique.
-Performed under reducing conditions.
-Predicted band size:25 kDa.

Research BackgroundTumor necrosis factor alpha (TNF-alpha), also known as TNF, TNFA or TNFSF2, is the prototypic cytokine of the TNF superfamily, and is a multifunctional molecule involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. Two receptors, TNF-R1 (TNF receptor type 1; CD120a; p55/60) and TNF-R2 (TNF receptor type 2; CD120b; p75/80), bind to TNF-alpha. TNF-alpha protein is produced mainly by macrophages, and large amounts of this cytokine are released in response to lipopolysaccharide, other bacterial products, and Interleukin-1 (IL-1). TNF-alpha is involved in fighting against the tumorigenesis, thus, is regarded as a molecular insight in cancer treatment.TNF-alpha Protein & AntibodyCancer ImmunotherapyImmune CheckpointImmunotherapyTargeted Therapy
