Verified Activity | 1. Western blot analysis of PRKAG3 on human PRKAG3 recombinant protein using anti-PRKAG3 antibody at 1/1,000 dilution. 2. Western blot analysis of PRKAG3 on HEK293 (1) and PRKAG3-hIgGFc transfected HEK293 (2) cell lysate using anti-PRKAG3 antibody at 1/1,000 dilution. 3. Immunohistochemical analysis of paraffin-embedded human rectum tissue using anti-PRKAG3 antibody. Counter stained with hematoxylin. 4. ICC staining PRKAG3 (green) and Actin filaments (red) in Hela cells. The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS. |

Research Background | AMP/ATP-binding subunit of AMP-activated protein kinase (AMPK), an energy sensor protein kinase that plays a key role in regulating cellular energy metabolism. In response to reduction of intracellular ATP levels, AMPK activates energy-producing pathways and inhibits energy-consuming processes: inhibits protein, carbohydrate and lipid biosynthesis, as well as cell growth and proliferation. AMPK acts via direct phosphorylation of metabolic enzymes, and by longer-term effects via phosphorylation of transcription regulators. Also acts as a regulator of cellular polarity by remodeling the actin cytoskeleton; probably by indirectly activating myosin.
