Western Blot -Positive WB detected in 293 whole cell lysate(treated with Calyculin A or Pervanadate) -All lanes Phospho-TP53 antibody at 1.28μg/ml -Secondary: Goat polyclonal to rabbit IgG at 1/50000 dilution -Predicted band size: 53 KDa -Observed band size: 53 KDa 2. Immunofluorescence staining of 293 cells(treated with 50mM Calyculin A for 30min) with TMAH-00976 at 1:100,counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG (H+L). |

Research Background | Acts as a tumor suppressor in many tumor types; induces growth arrest or apoptosis depending on the physiological circumstances and cell type. Involved in cell cycle regulation as a trans-activator that acts to negatively regulate cell division by controlling a set of genes required for this process. One of the activated genes is an inhibitor of cyclin-dependent kinases. Apoptosis induction seems to be mediated either by stimulation of BAX and FAS antigen expression, or by repression of Bcl-2 expression. Its pro-apoptotic activity is activated via its interaction with PPP1R13B/ASPP1 or TP53BP2/ASPP2.
