AGS-16C3F is an antibody-drug conjugate (ADC) targeting ENPP3 (also known as CD203c). This product consists of a humanized IgG1 monoclonal antibody against ENPP3 conjugated to the microtubule inhibitor monomethyl auristatin F (MMAF) via a non-cleavable linker. Description
AGS-16C3F is an antibody-drug conjugate (ADC) targeting ENPP3 (also known as CD203c). This product consists of a humanized IgG1 monoclonal antibody against ENPP3 conjugated to the microtubule inhibitor monomethyl auristatin F (MMAF) via a non-cleavable linker. AGS-16C3F functions by specifically binding to ENPP3 on the surface of tumor cells, undergoing internalization, and releasing MMAF intracellularly to induce cell cycle arrest and apoptosis. It exhibits potent anti-tumor and anti-proliferative activity in ENPP3-expressing metastatic renal cell carcinoma (mRCC) and systemic mastocytosis.
In vitro
AGS-16C3F inhibits proliferation of CD203c-high ROSA cells (96 h IC50: 1.1-2.73 nM), HMC-1.1 (IC50: 109.9 nM), and HMC-1.2 cells (IC50: 146.5 nM). It induces dose-dependent apoptosis (0.5-100 nM, 96-120 h) by disrupting microtubule dynamics [2].
In vivo
In pharmacological studies using NOD-SCID gamma mouse models xenografted with ROSA cells, systemic administration of AGS-16C3F (1 or 4 mg/kg; i.v.; once every 4 days for 4 doses) significantly reduces tumor burden. The treatment leads to a marked decrease in tumor cells within the peripheral blood, bone marrow, and spleen, while significantly prolonging survival without evidence of overt toxicity in complex physiological environments [2].
