CDKN2A germline mutations increase the risk of melanoma development. CDKN2A/CDK4 were the first high-penetrance melanoma genes identified. CDKN2A suppresses the initiation of melanoma invasion through inhibition of BRN2. It's a potential prognostic role of CDKN2A/B deletions in early disease stratification in pediatric B-ALL. CDKN2A coding region germline variants are associated with pancreatic adenocarcinoma (PC) susceptibility.
